The paper introduces Combocat, an innovative open-source platform designed to significantly speed up the discovery of synergistic drug combinations. By integrating acoustic liquid handling with an automated analytical pipeline, the system allows researchers to move beyond traditional, resource-heavy screening methods. The framework features a dense mode for high-fidelity mapping of drug interactions and a sparse mode that utilizes machine learning to predict full response landscapes from minimal experimental data. This predictive approach was validated through a massive screen of 9,045 combinations in neuroblastoma cells, marking a record for a single cell line. Ultimately, Combocat offers a scalable and user-friendly solution for identifying complex therapeutic interactions that can overcome drug resistance and reduce treatment toxicity.
