This research explores how DNA methylation, an epigenetic mechanism, drives the progression and clinical outcome of meningiomas, the most frequent primary brain tumor in adults. By analyzing large patient cohorts, the authors identified a specific hypermethylation signature that more accurately predicts tumor recurrence and aggressiveness than traditional histopathological grading. Their findings reveal that as these tumors evolve from benign to malignant states, they undergo long-range epigenetic silencing of clustered protocadherins. This molecular shift facilitates the nuclear translocation of β-catenin, a process that promotes tumor growth and suggests new targets for therapeutic intervention. Ultimately, the study advocates for integrating molecular profiling into clinical practice to better manage the diverse trajectories of the disease.
