The research introduces Hemin-His-Mn, a tumor-targeting nanozyme designed to overcome cancer immunotherapy resistance by triggering PANoptosis, a robust form of programmed cell death. This innovative nanomedicine acts by binding to the RIG-I protein, alleviating its autoinhibition to initiate PANoptosome assembly while simultaneously generating lethal reactive oxygen species to accelerate cell destruction. By inducing this immunogenic death, the platform effectively releases danger signals that reprogram the tumor microenvironment and activate a potent T cell-mediated immune response. Preclinical studies demonstrate that this dual-action strategy successfully shrinks tumors in mouse models and synergizes with immune checkpoint inhibitors to reverse treatment resistance. Ultimately, the study identifies RIG-I as a master sensor for PANoptosis and provides a clinically translatable modality for precision cancer immunotherapy.
