This research presents the development of a piezoelectric microneedle patch designed to treat melanoma by disrupting the harmful communication between the nervous and immune systems. The MT nanoparticles, composed of a manganese-doped metal-organic framework coated in neuron membranes, specifically target nociceptor neurons within the tumor microenvironment. When stimulated by ultrasound, these materials trigger the internalization of TRPV1 receptors and activate the STING pathway, which collectively reduces the release of immunosuppressive neuropeptides. By inhibiting these neural signals, the treatment restores anti-tumor immunity, enhances the activity of T cells and dendritic cells, and limits cancer metastasis. Experimental results in mice demonstrate that this minimally invasive strategy significantly suppresses tumor growth and works synergistically with existing immunotherapies to improve survival rates.
