This research identifies IL-17A-producing γδ T cells as a critical component of the immune defense against Mycobacterium abscessus, a difficult-to-treat non-tuberculous mycobacterial infection. Using mouse models and single-cell RNA sequencing, the study reveals that these specialized T cells are activated by macrophage-derived IL-1β and IL-23 via TLR2 signaling. Unlike many other mycobacterial responses, this pathway functions independently of interferon-gamma (IFNγ), offering a vital secondary defense when typical immune pathways are compromised by autoantibodies or genetic factors. The findings demonstrate that these γδ T cells directly eliminate bacteria through granzyme B-dependent cytotoxicity and clonal expansion of the Vγ6+ subset. Consequently, the research suggests that targeting and enhancing IL-17A+ γδ T cells could provide a promising therapeutic strategy for patients suffering from persistent or disseminated infections.
