The paper investigate how gut microbiota and tryptophan metabolism influence ischemic stroke severity through aryl hydrocarbon receptor (AHR) signaling in intestinal dendritic cells (DCs). Researchers discovered that indole-producing Escherichia coli worsens stroke outcomes by activating AHR in CD11c+ cells, whereas genetic deletion or pharmacological inhibition of AHR preserves conventional type 1 dendritic cells, reduces neuroinflammation, and promotes regulatory T cell (Treg) accumulation. Furthermore, experiments demonstrate that Tregs are essential for the neuroprotective benefits of AHR inhibition, highlighting a vital microbiota-gut-brain axis that can be targeted therapeutically.
