Adult glioma progression is heavily influenced by neural signaling, where excitatory circuits typically drive tumor growth through calcium dynamics. However, this study reveals that local inhibitory networks act as a natural brake on malignancy by forming functional GABAergic synapses with tumor cells. Enhancing inhibitory tone suppresses this calcium-driven oncogenic signaling, thereby reducing tumor proliferation and extending survival. Conversely, disrupting these inhibitory pathways accelerates cancer progression, demonstrating that brain tumor dynamics depend on the continuous balance between growth-promoting and growth-restraining neural inputs.
